In fluoresence microscopy the excitation light is reflected onto the sample by the dichroic, usually at an angle of incidence of 45°. The emitted fluoresence light is transmitted through the dichroic and onto the detector. It is crucial that the dichroic has good reflection and transmission characteristics and that the transition is a sharp as possible.
